Overview

Nab-sirolimus + bevacizumab is a combination therapy under investigation primarily for advanced or metastatic cancers, including colorectal cancer. Nab-sirolimus (ABI‑009, also called nab‑rapamycin) is a nanoparticle albumin-bound formulation of sirolimus, an mTOR inhibitor that blocks the mechanistic target of rapamycin kinase complex 1 (mTORC1), thereby inhibiting cell growth and proliferation. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis in tumors. The rationale behind combining these agents is to simultaneously suppress tumor cell proliferation via mTOR inhibition and reduce tumor blood supply through antiangiogenic effects. This regimen has been studied in phase I/II trials as first-line therapy in metastatic colorectal cancer and other solid tumors[1][4][6][8].
