nab-sirolimus + octreotide is an investigational combination therapy comprising nab-sirolimus, a nanoparticle albumin-bound formulation of sirolimus that inhibits the mammalian target of rapamycin (mTOR) pathway, with octreotide, a synthetic octapeptide analog of somatostatin that inhibits secretion of several hormones and exerts antiproliferative effects. nab-sirolimus delivers sirolimus with higher tumor accumulation and more effective mTOR signaling inhibition versus conventional mTOR inhibitors. Octreotide targets somatostatin receptors (especially SSTR2) and is often employed in the treatment of neuroendocrine tumors (NETs) for its hormone secretion-suppressing and antiproliferative properties. Preclinical data show additive antitumor and anti-proliferative effects for the combination in neuroendocrine tumor cell lines and xenograft models[2]. The combination is being explored in the treatment of advanced or metastatic neuroendocrine tumors.
