Nab-sirolimus (ABI-009, also known as nab-rapamycin) is a nanoparticle albumin-bound formulation of sirolimus developed to enhance drug delivery and accumulation in solid tumors. It acts as a potent inhibitor of the mechanistic target of rapamycin (mTOR), a key regulator of cell growth and proliferation. Temozolomide is an oral alkylating agent that induces DNA damage leading to tumor cell death. Radiotherapy uses ionizing radiation to kill cancer cells or inhibit their growth. The combination regimen has been investigated primarily for pediatric and adult solid tumors, including gliomas, with the aim of improving efficacy through synergistic mechanisms—nab-sirolimus targeting mTOR signaling pathways, temozolomide causing DNA damage, and radiotherapy providing local tumor control[1][2][3]. Nab-sirolimus was developed by Aadi Bioscience.
