**nacubactam + meropenem** is a fixed-dose intravenous combination of **nacubactam**, a novel diazabicyclooctane non-β-lactam β-lactamase inhibitor, and **meropenem**, a broad-spectrum carbapenem antibiotic. - **Nacubactam** inhibits bacterial serine β-lactamases (classes A and C, some D, e.g., KPC, OXA, CTX-M, SHV, TEM), restoring and potentiating β-lactam efficacy against multidrug-resistant Gram-negative bacteria. - Nacubactam also directly inhibits penicillin-binding protein 2 (PBP2) in Enterobacteriaceae, providing intrinsic antibacterial activity. - **Meropenem** disrupts bacterial cell wall synthesis by binding to PBPs and is active against a range of Gram-negative pathogens, including carbapenem-resistant strains. - The combination enhances antibacterial activity, including against ceftazidime-avibactam-resistant and carbapenemase-producing *Enterobacteriaceae*, such as *Klebsiella pneumoniae* and *Escherichia coli*, and is being developed primarily for complicated urinary tract infections (cUTIs) and other difficult-to-treat, multidrug-resistant infections[1][2][3][4][5][6][7][8]. - Developed for intravenous administration in hospitalized adult patients, it offers improved efficacy over meropenem alone against resistant isolates.
