Drug intelligence / Profile preview
nadunolimab + azacitidine (CAN04 + azacitidine) is an investigational combination regimen consisting of nadunolimab (CAN04), a fully humanized monoclonal antibody targeting interleukin-1 receptor accessory protein (IL1RAP), and azacitidine, a small-molecule hypomethylating chemotherapy agent. Developed by Cantargia AB, nadunolimab works by blocking IL-1α and IL-1β signaling and inducing antibody-dependent cellular cytotoxicity (ADCC) against IL1RAP-expressing cells, such as leukemia stem cells. Azacitidine acts as an antimetabolite and epigenetic modulator. The combination is being evaluated in clinical trials, including an investigator-initiated Phase 1b/2a trial at MD Anderson Cancer Center, for the treatment of high-risk myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML).
