Overview

Nadunolimab + toripalimab is an investigational combination immunotherapy consisting of an anti-IL-1RAP monoclonal antibody (nadunolimab) and an anti-PD-1 monoclonal antibody (toripalimab). This combination is specifically being evaluated for the treatment of chemotherapy-refractory metastatic microsatellite stable (MSS) colorectal cancer, a population that typically shows poor response to single-agent checkpoint inhibitors. Nadunolimab targets the interleukin-1 receptor accessory protein (IL-1RAP), which is overexpressed on various cancer cells and immune cells, thereby inhibiting IL-1 signaling and potentially reducing tumor-promoting inflammation. Toripalimab blocks the PD-1 receptor on T-cells, preventing its interaction with PD-L1/PD-L2 and restoring the immune system's ability to recognize and attack tumor cells. By combining these two agents, the therapy aims to overcome the immunosuppressive environment typical of MSS colorectal cancer.
