Nafabelltan (CKD-314) is a synthetic small molecule serine protease inhibitor developed by Chong Kun Dang. It is closely related to nafamostat and has been investigated primarily for the treatment of COVID-19 pneumonia. Nafabelltan acts by inhibiting a range of serine proteases involved in coagulation and inflammation, including thrombin, plasmin, kallikrein, trypsin, complement system proteases (Cl esterase), and coagulation factors VIIa, Xa, and XIIa. It also inhibits transmembrane protease serine 2 (TMPRSS2), which plays a key role in viral entry for coronaviruses such as SARS-CoV-2. This mechanism underlies its potential antiviral activity against COVID-19[5][7][9]. Nafabelltan has anticoagulant and anti-inflammatory properties as well.
