Naltrexone + methylphenidate is an investigational drug combination consisting of naltrexone, an opioid receptor antagonist, and methylphenidate, a central nervous system stimulant. The combination is evaluated primarily for the treatment of attention-deficit/hyperactivity disorder (ADHD). Naltrexone blocks μ-opioid receptors, reducing the euphoric effects and potentially the abuse liability of methylphenidate, while methylphenidate acts as a stimulant to improve ADHD symptoms. Clinical studies found the combination did not interfere with the clinical efficacy of methylphenidate for ADHD and showed no increase in adverse events, and also attenuated stimulant-induced euphoria in adults with ADHD[1][3][5][6].
