Namilumab + methotrexate is an investigational combination regimen for rheumatoid arthritis, in which namilumab, a fully human IgG1 monoclonal antibody targeting granulocyte-macrophage colony-stimulating factor (GM-CSF), is administered alongside methotrexate. Namilumab blocks GM-CSF signaling, reducing inflammation relevant to rheumatoid arthritis pathogenesis, while methotrexate is a standard disease-modifying anti-rheumatic drug (DMARD) that inhibits folate metabolism and suppresses immune functions. Clinical studies, including phase 1b and phase 2 trials, have evaluated this combination in patients with moderate to severe rheumatoid arthritis inadequately controlled on methotrexate alone, reporting signs of efficacy and a generally acceptable safety profile.
