Nanoxel + Herzuma is a combination chemotherapy regimen being evaluated for the treatment of HER2-positive metastatic salivary duct carcinoma. The regimen consists of Nanoxel, a polymeric micelle formulation of the taxane paclitaxel, and Herzuma (trastuzumab-pkrb), a biosimilar monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2). Paclitaxel works by binding to tubulin and stabilizing microtubules, which inhibits mitosis and leads to cell death. Herzuma binds to the extracellular domain of the HER2 receptor, inhibiting downstream signaling pathways and recruiting immune effector cells to mediate antibody-dependent cellular cytotoxicity (ADCC). This combination is being studied in a Phase II clinical trial led by Samsung Medical Center to address the high prevalence of HER2 overexpression in salivary duct carcinomas, a rare and aggressive malignancy.
