**Nanrilkefusp alfa + pembrolizumab** is an investigational combination therapy for advanced solid tumors. Nanrilkefusp alfa is a recombinant fusion protein that acts as an interleukin-15 receptor βγ (IL-15Rβγ) superagonist, designed to selectively stimulate cytotoxic natural killer (NK) and CD8+ T cells and promote an anti-tumor immune response. Pembrolizumab is a monoclonal antibody checkpoint inhibitor targeting programmed cell death protein 1 (PD-1), enhancing T cell anti-tumor activity by blocking inhibitory signaling. This combination aims to boost both innate and adaptive immune responses for synergistic anti-tumor effects. Development of this combination for solid tumors was discontinued due to insufficient efficacy in clinical trials, but data suggest a favorable safety profile and evidence of immune activation in some checkpoint-resistant/refractory patients[1][4][9][10].
