This is a combination regimen comprised of **napabucasin**, **fluorouracil (5-FU)**, **oxaliplatin**, **leucovorin**, and **bevacizumab**. Napabucasin is an orally bioavailable agent that is activated by NAD(P)H:quinone oxidoreductase 1, causing reactive oxygen species generation and hypothesized to modulate multiple oncogenic pathways, particularly STAT3-driven signaling, resulting in potential cancer cell death. Fluorouracil is a pyrimidine analog that inhibits thymidylate synthase, disrupting DNA synthesis. Oxaliplatin is a platinum-based alkylating agent that inhibits DNA replication. Leucovorin is used to potentiate the efficacy of fluorouracil. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A, inhibiting angiogenesis crucial for tumor growth. This combination is under investigation as an antineoplastic regimen (especially for metastatic colorectal cancer) designed to target angiogenesis, DNA synthesis/repair, and cancer stemness.
