**Napabucasin + nivolumab** is an experimental combination therapy evaluated in clinical trials primarily for cancer treatment. Napabucasin is a small molecule that acts as a cancer stemness inhibitor, mainly by targeting and inhibiting the activity of signal transducer and activator of transcription 3 (STAT3) driven gene transcription, thereby reducing cancer cell "stemness" and resistance to standard therapies. Nivolumab is a fully human monoclonal antibody that blocks the programmed death-1 (PD-1) receptor, thereby enhancing T-cell immune responses against tumor cells. This combination is being studied to potentially harness the complementary mechanisms of targeting cancer stemness (napabucasin) and activating immune-mediated tumor cell killing (nivolumab), particularly in solid tumors and advanced malignancies.
