Overview

Napabucasin + sorafenib is an investigational **oral combination therapy** evaluated primarily for the treatment of unresectable or advanced hepatocellular carcinoma (HCC). Napabucasin is a small molecule that acts as a bioactivatable agent targeting NAD(P)H:quinone oxidoreductase 1, leading to inhibition of signal transducer and activator of transcription 3 (STAT3) driven pathways and cancer cell stemness. Sorafenib is a multikinase inhibitor targeting RAF kinases and various receptor tyrosine kinases involved in tumor cell proliferation and angiogenesis. The combination has been studied in phase I and II clinical trials for safety, tolerability, pharmacokinetics, and preliminary efficacy in HCC, showing manageable adverse events and disease stabilization in a subset of patients[1][2][3].
