Overview

Naphthoquine + azithromycin is a fixed-dose combination drug formulated in a 1:1 ratio for the prophylaxis of malaria. Naphthoquine is an antimalarial agent with a long elimination half-life (>20 days), providing extended protection against Plasmodium species. Azithromycin is a macrolide antibiotic that also exhibits delayed antimalarial effects by targeting the apicoplast of Plasmodium parasites. The combination has demonstrated high efficacy as monthly single-dose prophylaxis, offering over 90% protective efficacy against all malaria species within one month and complete protection against relapsing malaria caused by Plasmodium vivax and Plasmodium ovale. Both agents have favorable safety profiles; transient elevation of liver aminotransferases was observed but not statistically significant compared to placebo[1][3]. The synergistic interaction between the two drugs enhances their antimalarial activity[2][4].
