Overview

Naproxen + rabeprazole is a fixed-dose combination of two small molecule drugs used to manage pain and inflammation while reducing the risk of gastrointestinal side effects. Naproxen is a nonsteroidal anti-inflammatory drug (NSAID) that works by inhibiting cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2), enzymes responsible for prostaglandin synthesis, thereby providing analgesic, anti-inflammatory, and antipyretic effects[5]. Rabeprazole is a proton pump inhibitor that irreversibly inhibits the hydrogen-potassium ATPase enzyme system (proton pump) in gastric parietal cells, leading to suppression of gastric acid secretion[3][6]. The combination is intended for patients who require NSAID therapy but are at increased risk for NSAID-induced gastric ulcers or gastrointestinal complications. This type of combination has been marketed with other proton pump inhibitors (e.g., esomeprazole), but specific fixed-dose combinations with rabeprazole may be available in some regions.
