Overview

This is a fixed-dose combination of three drugs: - **Naproxen** is a nonsteroidal anti-inflammatory drug (NSAID) that works by inhibiting cyclooxygenase enzymes, thereby reducing the synthesis of prostaglandins involved in pain and inflammation. - **Rebamipide** is a gastroprotective agent that enhances mucosal defense, promotes healing, and reduces gastrointestinal injury. Its mechanisms include increasing mucus production, enhancing blood flow to the gastric mucosa, scavenging free radicals, upregulating prostaglandin synthesis (PGE2 and PGI2), suppressing inflammatory cytokines such as TNF-α, and promoting epithelial repair through increased expression of epidermal growth factor[1][4]. - **Rabeprazole** is a proton pump inhibitor (PPI) that irreversibly inhibits the H+/K+ ATPase enzyme in gastric parietal cells. This action suppresses both basal and stimulated gastric acid secretion[6][8]. The combination aims to provide effective anti-inflammatory therapy while minimizing NSAID-induced gastrointestinal damage by combining an NSAID with agents that protect the GI tract. Clinical trials have evaluated this triple therapy for prevention of naproxen-induced gastric lesions[5][2]. The primary indications are for patients requiring NSAIDs who are at risk for GI complications.
