Overview

Naptumomab estafenatox + interferon alpha is a combination immunotherapy investigated primarily for advanced and metastatic renal cell carcinoma. **Naptumomab estafenatox** is a fusion protein: the Fab fragment targets the tumor-associated antigen 5T4 on cancer cells, while the fused staphylococcal enterotoxin (superantigen SEA/E-120, engineered for reduced MHC class II and antibody binding) activates and redirects T cells to attack 5T4-positive tumor cells, leading to cytokine release and antitumor effects. **Interferon alpha** is a cytokine that enhances immune system activity against cancer. This combination was tested in a phase II/III trial versus interferon alpha alone. The combination did not show a statistically significant increase in overall survival compared to interferon alpha alone in the general population, but exploratory subgroup analysis suggested benefit in patients with low baseline anti-SEA/E-120 antibody and IL6 levels.
