Overview

**Narlaprevir + ritonavir + raltegravir** is a combination regimen being investigated for the management of HIV-1 infection, particularly in patients co-infected with hepatitis C virus (HCV). Narlaprevir is a hepatitis C virus NS3 protease inhibitor, ritonavir is an HIV-1 protease inhibitor functioning here primarily as a pharmacokinetic booster (enhancing plasma concentrations of other protease inhibitors by inhibiting CYP3A4), and raltegravir is an HIV-1 integrase strand transfer inhibitor (INSTI) that inhibits the integration of viral DNA into the host genome. The combination is studied to evaluate pharmacokinetic interactions and potential dual regimens that spare nucleoside/nucleotide reverse transcriptase inhibitors (NRTIs) due to their long-term toxicities. This regimen is under clinical investigation, with no evidence of approval for standard therapy as of now.
