Narmafotinib + Olomorasib is an investigational small molecule combination therapy being developed by Amplia Therapeutics in collaboration with Eli Lilly. The combination pairs Amplia's highly potent and selective focal adhesion kinase (FAK) inhibitor, narmafotinib (AMP945), with Eli Lilly's next-generation, covalent KRAS G12C inhibitor, olomorasib (LY3537982). FAK activation has been identified as a key mediator of adaptive resistance to KRAS G12C inhibitors, promoting tumor cell survival and remodeling the tumor microenvironment. By combining FAK and KRAS G12C inhibition, this dual-targeted approach aims to overcome resistance mechanisms, thereby deepening and prolonging therapeutic responses. The combination is being evaluated in a Phase 1b/2b clinical trial as a second-line treatment for patients with advanced KRAS G12C-mutated non-small cell lung cancer (NSCLC).
