Nateglinide + metformin is an oral combination therapy used to improve glycemic control in adults with type 2 diabetes mellitus. This fixed-dose combination brings together two complementary antidiabetic agents from different classes. Nateglinide, a meglitinide, stimulates insulin secretion from pancreatic beta cells in response to meals, thereby reducing postprandial blood glucose levels. Metformin, a biguanide, primarily decreases hepatic glucose production and increases insulin sensitivity in peripheral tissues, leading to reduced fasting plasma glucose. The combination is indicated when monotherapy with either agent does not provide adequate glycemic control or for initial therapy in certain patients.
