navarixin + pembrolizumab

Navarixin + pembrolizumab is a combination therapy consisting of navarixin, an oral small molecule antagonist of C-X-C motif chemokine receptor 2 (CXCR2), and pembrolizumab, an intravenous monoclonal antibody targeting programmed cell death protein 1 (PD-1). Navarixin inhibits CXCR2, which plays a role in tumor progression and immune evasion by modulating neutrophil recruitment and the tumor microenvironment. Pembrolizumab blocks PD-1 to enhance T-cell mediated anti-tumor immunity. The combination was investigated for potential synergistic effects in advanced or metastatic solid tumors including castration-resistant prostate cancer (CRPC), microsatellite-stable colorectal cancer (MSS CRC), and non-small-cell lung cancer (NSCLC). In phase 2 clinical trials, the combination did not demonstrate sufficient efficacy but showed manageable safety and tolerability profiles[1][2][4].
