**Navitoclax + docetaxel** is an investigational combination therapy consisting of navitoclax, an orally available small-molecule inhibitor of Bcl-2 family anti-apoptotic proteins (notably Bcl-2, Bcl-xL, and Bcl-w), and docetaxel, a cytotoxic taxane chemotherapy agent that promotes microtubule stabilization and subsequent cell cycle arrest. Navitoclax sensitizes tumor cells to apoptosis by inhibiting the anti-apoptotic Bcl-2 family proteins, which are often upregulated in cancer and contribute to chemoresistance, while docetaxel induces mitotic arrest and cytotoxicity via microtubule stabilization. The combination is primarily being studied in patients with advanced solid tumors, including non-small cell lung cancer (NSCLC), where preclinical and clinical studies have shown potential for enhanced antitumor activity compared to either agent alone. The combination is still under investigation; clinical studies have noted dose-limiting toxicities including myelosuppression, thrombocytopenia, fatigue, neutropenia, and gastrointestinal effects, but also observed partial responses and clinical benefit in some patients[1][3][5][7].
