Nazartinib + trametinib is an investigational combination therapy consisting of two small molecule inhibitors targeting distinct pathways involved in cancer cell growth and survival. Nazartinib (EGF816) is a third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor designed to target activating EGFR mutations and overcome resistance mechanisms in non-small cell lung cancer (NSCLC). Trametinib is a selective inhibitor of mitogen-activated protein kinase kinase (MEK), part of the RAS/RAF/MEK/ERK signaling pathway. The rationale for combining these agents stems from evidence that co-activation of the RAS/MEK pathway can mediate resistance to EGFR inhibition; thus, dual blockade may restore or enhance sensitivity to EGFR-targeted therapies. This combination has been studied in early-phase clinical trials for patients with advanced NSCLC harboring EGFR mutations[1][3].
