NBTXR3 + cisplatin is a combination therapy under investigation for locally advanced head and neck squamous cell carcinoma (LA-HNSCC) and other locally advanced solid tumors. **NBTXR3** is a first-in-class intratumoral **radioenhancer** composed of functionalized **hafnium oxide nanoparticles**, designed to be injected directly into tumors. Upon exposure to ionizing radiation, these nanoparticles amplify the radiation dose deposited inside tumor cells, increasing cytotoxicity via elevated reactive oxygen species generation and tumor cell death, while sparing surrounding normal tissue. This radioenhancing effect can boost the efficacy of radiotherapy. **Cisplatin** is a well-established small molecule chemotherapeutic agent that acts as a DNA crosslinker, leading to tumor cell apoptosis. The combination is tested primarily as part of chemoradiation (concomitant chemoradiotherapy), using NBTXR3 intratumoral injection with standard-of-care low-dose weekly cisplatin and radiotherapy in patients with locally advanced or recurrent disease. NBTXR3 is developed by Nanobiotix.
