NC318 + pemetrexed + carboplatin is an investigational combination regimen for cancer therapy. NC318 is a monoclonal antibody targeting Siglec-15 (Sialic acid-binding immunoglobulin-type lectin 15), an immune suppressor upregulated in various cancers and thought to play a role in immune evasion of tumor cells. Pemetrexed is an antifolate chemotherapeutic that inhibits multiple folate-dependent enzymes involved in purine and pyrimidine synthesis, suppressing DNA and RNA synthesis in rapidly dividing cells. Carboplatin is a platinum-based chemotherapeutic that induces DNA crosslinks, leading to apoptosis of cancer cells. Pemetrexed and carboplatin are commonly used together as first-line chemotherapy for non-small cell lung cancer (NSCLC) and mesothelioma. The addition of NC318 is under clinical investigation to determine whether targeting Siglec-15 can improve antitumor immune responses in combination with standard chemotherapy.
