Overview

NC410 + pembrolizumab is an investigational combination therapy designed for advanced or metastatic solid tumors, particularly those that are immune checkpoint inhibitor (ICI) naïve or refractory, including microsatellite stable (MSS)/microsatellite instability-low (MSI-L) colorectal cancer (CRC) and ovarian cancer. NC410 is a dimeric LAIR-2 fusion protein (LAIR-2-IgG1 Fc) that antagonizes collagen-mediated immunosuppression in the tumor extracellular matrix by competing with LAIR-1, an inhibitory receptor on immune cells. This remodeling of the ECM enables greater immune cell infiltration. Pembrolizumab (Keytruda) is a well-established monoclonal antibody targeting the PD-1 immune checkpoint, stimulating the anti-tumor immune response. The combination aims to increase immune cell accessibility to tumors while simultaneously reducing immunosuppression and unleashing immune activity against cancer cells. Developed for advanced unresectable/metastatic MSS/MSI-low CRC and ovarian cancer, with some investigation in other advanced solid tumors. Preclinical and clinical evidence shows this combination is promising for hard-to-treat cancers with dense ECM and low response rates to standard immunotherapies[2][4].
