NDI-101150 + pembrolizumab is an investigational combination therapy being studied in advanced solid tumors, notably renal cell carcinoma and other metastatic or relapsed cancers. NDI-101150 is an oral, small molecule, highly selective inhibitor of hematopoietic progenitor kinase 1 (HPK1), an intracellular negative regulator of T-cell, B-cell, and dendritic cell immune responses. By inhibiting HPK1, NDI-101150 aims to enhance antitumor immune responses. Pembrolizumab is a monoclonal antibody that targets PD-1 on lymphocytes and blocks its interaction with PD-L1/PD-L2, thereby promoting immune-mediated tumor cell killing. The combination is designed to work synergistically: NDI-101150 relieves T cell inhibition at the intracellular signaling level, while pembrolizumab blocks the PD-1 immune checkpoint on the cell surface. This approach is currently being evaluated in Phase 1/2 trials for safety, tolerability, and preliminary efficacy[1][3][5][7][9].
