Overview
Nectin-4 ADC (NMSCMS) is an investigational antibody-drug conjugate (ADC) developed by Nerviano Medical Sciences (NMS Group). The drug targets Nectin-4 (Poliovirus receptor-related protein 4), a cell surface protein that is highly expressed in several solid tumors, including bladder, breast, and lung cancers, while having limited expression in normal tissues. This ADC utilizes NMS's proprietary platform to conjugate a monoclonal antibody to a next-generation anthracycline-derived cytotoxic payload. The mechanism of action involves the selective binding of the ADC to Nectin-4 on tumor cells, followed by internalization and the intracellular release of the anthracycline payload. The released payload then exerts its anti-tumor effect through DNA intercalation and the inhibition of topoisomerase II, leading to DNA damage and apoptosis. The program is currently in the preclinical stage of development.
