Overview

Nedaplatin + capecitabine is a combination chemotherapy regimen primarily utilized in the treatment of solid tumors, most notably esophageal squamous cell carcinoma and gastric cancer. Nedaplatin is a second-generation platinum analogue that induces cytotoxicity by forming DNA adducts and cross-links, inhibiting DNA replication and transcription. Capecitabine is an orally administered fluoropyrimidine carbamate that acts as a prodrug, undergoing a three-step enzymatic conversion to 5-fluorouracil (5-FU) within the body, with the final conversion occurring preferentially in tumor tissue via thymidine phosphorylase. Once converted, 5-FU inhibits thymidylate synthase and incorporates into RNA and DNA, leading to cell death. This combination is often investigated as a more convenient, oral-inclusive alternative to the traditional cisplatin and intravenous 5-fluorouracil (PF) regimen, showing comparable efficacy with a distinct safety profile, often characterized by reduced gastrointestinal and renal toxicity but increased myelosuppression.
