Nedaplatin + gemcitabine is a combination chemotherapy regimen consisting of nedaplatin, a platinum-based antineoplastic agent, and gemcitabine, a nucleoside analog metabolic inhibitor. Nedaplatin acts primarily by forming DNA crosslinks that inhibit DNA replication and transcription, leading to apoptosis in rapidly dividing cells. Gemcitabine is incorporated into DNA during replication as a cytidine analog, causing chain termination and also inhibiting ribonucleotide reductase, further depleting deoxynucleotide pools required for DNA synthesis. This combination has been investigated primarily for the treatment of advanced non-small cell lung cancer (NSCLC) and head and neck squamous cell carcinoma (HNSCC), showing acceptable efficacy and safety profiles in phase I/II trials[1][2][3]. The regimen is typically administered intravenously every three weeks.
