This is a combination regimen consisting of three agents: - **Nedaplatin** is a second-generation platinum-based chemotherapeutic agent that forms DNA crosslinks, inhibiting DNA replication and transcription, leading to apoptosis in rapidly dividing tumor cells. It has demonstrated efficacy with relatively lower nephrotoxicity compared to cisplatin and is used primarily in the treatment of various solid tumors, including non-small cell lung cancer (NSCLC), esophageal squamous cell carcinoma (ESCC), cervical cancer, and urothelial cancers[1][2][3][4][5][6]. - **Paclitaxel** is a taxane-class chemotherapeutic that stabilizes microtubules and prevents their depolymerization during cell division, resulting in mitotic arrest and apoptosis. It is widely used for multiple malignancies including NSCLC, ESCC, cervical cancer, ovarian cancer, breast cancer[1][2][3][5][6]. - **Sintilimab** (also known as Tyvyt) is a monoclonal antibody targeting programmed cell death protein 1 (PD-1). By blocking PD-1 interaction with its ligands PD-L1/PD-L2 on tumor cells or antigen-presenting cells, sintilimab restores T-cell mediated anti-tumor immunity. Sintilimab has been approved for relapsed/refractory classical Hodgkin lymphoma in China and investigated for other solid tumors such as NSCLC[7]. The combination of nedaplatin plus paclitaxel has shown promising activity as first-line therapy for advanced/metastatic NSCLC and ESCC with manageable toxicity profiles[1][2][3][4][5]. Adding an immune checkpoint inhibitor like sintilimab may further enhance anti-tumor efficacy by combining cytotoxic chemotherapy with immunotherapy.
