Nenocorilant + nivolumab is a combination therapy consisting of nenocorilant (CORT118335), a selective glucocorticoid receptor (GR) modulator, and nivolumab, a programmed death-1 (PD-1) blocking monoclonal antibody. Developed by Corcept Therapeutics, this combination is designed to overcome immune resistance in solid tumors. Glucocorticoid receptor activation in the tumor microenvironment can suppress immune responses and promote tumor growth; by modulating GR, nenocorilant aims to enhance the anti-tumor activity of checkpoint inhibitors like nivolumab. The combination is currently being evaluated in the Phase 2 SYNERGY study for patients with advanced solid tumors, including platinum-resistant ovarian cancer.
