NEO-201 + pembrolizumab is a combination therapy under clinical investigation for the treatment of advanced solid tumors, particularly in patients who have progressed on prior checkpoint inhibitor therapies. NEO-201 is a humanized IgG1 monoclonal antibody developed to target tumor-associated antigens, specifically core 1 O-glycans, which are overexpressed on certain cancer cells and immunosuppressive cells such as granulocytic myeloid-derived suppressor cells (gMDSCs) and regulatory T cells (Tregs). Its mechanisms include direct tumor cell killing via antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and blockade of the CEACAM5/CEACAM1 immune checkpoint inhibitory pathway. Pembrolizumab is an anti-PD-1 immune checkpoint inhibitor that restores T-cell mediated antitumor activity. The combination aims to overcome resistance to checkpoint inhibitors by depleting immunosuppressive cells in the tumor microenvironment and reactivating immune responses against cancer[3][5][6][8].
