Overview

The Neo-DC vaccine is a personalized, autologous dendritic cell-based immunotherapy developed by Fudan University for the treatment of advanced solid tumors, including head and neck cancer, non-small cell lung cancer (NSCLC), and esophageal squamous cell carcinoma (ESCC). The therapeutic process involves identifying unique tumor neoantigens through genomic sequencing of a patient's tumor, synthesizing these neoantigens as peptides, and using them to pulse the patient's own harvested dendritic cells. Once re-infused, these mature, neoantigen-presenting dendritic cells activate and expand tumor-specific T cells, particularly CD8+ cytotoxic T lymphocytes, to recognize and eliminate cancer cells expressing those specific mutations. This approach aims to overcome the low immunogenicity of many tumors by focusing the immune system on highly specific, non-self targets.
