Overview

Neo-Vac-Mn + toripalimab is an investigational combination immunotherapy regimen being developed by Xiangya Hospital of Central South University for the treatment of metastatic cutaneous melanoma. The therapy consists of Neo-Vac-Mn, a personalized cancer vaccine composed of patient-specific neoantigen peptides identified through tumor sequencing, and toripalimab, a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. The vaccine is designed to prime the immune system to recognize and attack tumor cells expressing specific mutations, while toripalimab acts as a checkpoint inhibitor to prevent the tumor from evading the immune response by blocking the PD-1/PD-L1 pathway. The regimen also incorporates adjuvants such as recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF) and topical imiquimod, a Toll-like receptor 7 (TLR7) agonist, to enhance the dendritic cell-mediated presentation of the neoantigens and boost the overall anti-tumor immune response.
