Overview

**NEO212 + pembrolizumab** is an investigational combination therapy consisting of NEO212, a novel orally administered small-molecule chemotherapeutic conjugate of temozolomide and perillyl alcohol, and pembrolizumab, a well-established humanized monoclonal antibody that targets the PD-1 immune checkpoint receptor. NEO212 is under evaluation for its ability to cross the blood-brain barrier and exert antitumor activity in brain tumors and metastases. Pembrolizumab acts by blocking the PD-1 receptor on T-cells, leading to enhanced immune activation against tumor cells. The combination is being tested in phase 1/2 clinical trials for safety, pharmacokinetics, and efficacy in solid tumors with uncontrolled brain metastases, including various primary cancers such as melanoma, non-small cell lung cancer, breast cancer, and others.
