A personalized DNA-based cancer vaccine platform developed by Washington University School of Medicine. The vaccine consists of closed circular DNA plasmids (pING vector) that encode patient-specific tumor neoantigens identified through exome sequencing of the individual's tumor. To enhance the immune response, the polyepitope sequences are fused to a mutated ubiquitin (G76V) at the N-terminus, which promotes proteasomal degradation and subsequent HLA class I and II presentation. The vaccine is administered as naked DNA via intramuscular electroporation using an integrated device to facilitate cellular uptake. It is primarily investigated in the adjuvant setting for patients with triple-negative breast cancer and other solid tumors to prevent recurrence by stimulating a robust T-cell mediated anti-tumor response.
