This personalized neoantigen peptide vaccine is a biologic therapeutic developed by the Washington University School of Medicine for the treatment of pancreatic adenocarcinoma. The vaccine is custom-manufactured for each patient using synthetic long peptides (SLPs) that incorporate prioritized neoantigens identified through exome sequencing of the patient's resected tumor tissue, along with personalized mesothelin epitopes. It is co-administered with poly-ICLC, a toll-like receptor 3 (TLR3) agonist that acts as a molecular adjuvant to enhance the immune response. The vaccine is designed to generate robust neoantigen-specific CD4+ and CD8+ T cell responses to eliminate residual cancer cells following surgical resection and adjuvant chemotherapy, and is currently being evaluated in Phase 1 clinical trials (NCT03956056).
