NeoPep Vaccine1 + Poly-ICLC is a combination immunotherapy under investigation for the treatment of glioblastoma and other cancers. NeoPep Vaccine1 (NPVAC1) consists of up to five synthetic peptides derived from human cytomegalovirus (HCMV) antigens, designed to stimulate an immune response against tumor cells. It is administered in combination with Poly-ICLC, a stabilized form of polyinosinic-polycytidylic acid that acts as an immune adjuvant by mimicking viral double-stranded RNA and activating innate immunity through Toll-like receptor 3 (TLR3) and melanoma differentiation-associated protein 5 (MDA5). This activation leads to the production of type I interferons and pro-inflammatory cytokines, enhancing antigen presentation and promoting T-cell mediated anti-tumor responses. The combination aims to induce robust tumor-specific T cell populations in patients with newly diagnosed glioblastoma[5][6][7].
