NeoPep Vaccine2 + Poly-ICLC is a combination immunotherapy consisting of a personalized neoantigen peptide vaccine (NeoPep Vaccine2 or NPVAC2) and the immunomodulatory adjuvant polyinosinic-polycytidylic acid stabilized with poly-L-lysine and carboxymethylcellulose (Poly-ICLC; also known as Hiltonol). NeoPep Vaccine2 is composed of up to 20 de novo synthesized peptides tailored for each patient based on their tumor-specific mutanome. These peptides are designed to elicit a robust T-cell immune response against cancer cells expressing the corresponding mutated antigens. Poly-ICLC acts as an immune adjuvant by mimicking double-stranded RNA and activating Toll-like receptor 3 (TLR3), thereby enhancing dendritic cell maturation, type I interferon production, and overall anti-tumor immunity. This combination is under investigation primarily for glioblastoma but may be relevant in other cancers where personalized neoantigen vaccines are being explored[4][5][6].
