Overview

**Neosaxitoxin + bupivacaine + epinephrine** is an investigational combination for local anesthesia designed to produce prolonged and potent sensory and pain blockade. Neosaxitoxin is a site 1 sodium channel blocker (saxitoxin analog), bupivacaine is an amino amide local anesthetic, and epinephrine acts as a vasoconstrictor to reduce systemic absorption and toxicity. In preclinical and Phase 1 studies, the combination leads to significantly prolonged duration and intensity of sensory blockade compared to individual agents or their binary combinations, with improved safety and reduced systemic adverse effects.
