Overview

NEP(1-40) is a synthetic peptide comprising the first 40 amino acids of the Nogo-66 inhibitory domain of the myelin-associated protein Nogo-A. It functions as a competitive antagonist of the Nogo-66 receptor (NgR1, also known as RTN4R), which is expressed on the surface of axons. By blocking the interaction between Nogo-66 and NgR1, NEP(1-40) prevents the activation of downstream signaling pathways, such as the Rho/ROCK cascade, that inhibit neurite outgrowth and axonal regeneration in the central nervous system. This peptide has been extensively studied in preclinical models of spinal cord injury (SCI) and traumatic brain injury (TBI) to promote nerve repair. Additionally, recent research has explored its role in mitigating hyperalgesia and neural injury in sickle cell disease by inhibiting Nogo-A-mediated pathways.
