Neratinib + temsirolimus is a combination of two small molecule drugs used in oncology research. Neratinib is an irreversible pan-HER tyrosine kinase inhibitor that targets the human epidermal growth factor receptor (HER) family—including HER1 (EGFR), HER2 (ErbB2), and HER4—blocking downstream signaling pathways involved in tumor cell proliferation and survival. Temsirolimus is an inhibitor of mammalian target of rapamycin (mTOR), a key regulator of cell growth and metabolism. The rationale for combining these agents stems from preclinical evidence showing synergy between HER2 inhibition and mTOR inhibition in various cancer models. This combination has been evaluated in phase I clinical trials for patients with advanced solid tumors, including breast cancer resistant to trastuzumab and non-small-cell lung cancer with HER2 mutations. The regimen demonstrated tolerability at specific dose levels and showed antitumor activity across multiple tumor types[1][2][4].
