Neratinib + vinorelbine is a combination therapy consisting of neratinib, an irreversible pan-HER tyrosine kinase inhibitor, and vinorelbine, a chemotherapeutic agent. This combination is primarily used in the treatment of HER2-positive metastatic breast cancer, particularly in patients who have been previously treated with anti-HER2 therapy. Neratinib works by irreversibly binding to and inhibiting HER1, HER2, and HER4 receptors, preventing receptor phosphorylation and blocking downstream signaling through the MAPK and AKT pathways. The combination has shown promising antitumor activity in clinical trials, with an objective response rate of 41% in patients who had not received prior lapatinib treatment.
