Overview

Nesvacumab + aflibercept is a fixed-dose combination of two monoclonal antibodies administered as a single co-formulated intravitreal injection. Nesvacumab is an anti-angiopoietin-2 (Ang2) antibody, while aflibercept is an anti-vascular endothelial growth factor (VEGF) agent. The combination was developed to target both Ang2 and VEGF pathways, which are implicated in pathological ocular neovascularization and increased vascular permeability seen in diseases such as neovascular age-related macular degeneration (nAMD) and diabetic macular edema (DME). Clinical studies aimed to determine if dual inhibition would provide superior efficacy over VEGF inhibition alone. However, phase 2 trials found no significant additional benefit in visual acuity or anatomical outcomes compared to aflibercept monotherapy[1][3][4][6][8].
