Both drugs act by inhibiting the reverse transcriptase enzyme, which is essential for the replication of HIV. The combination can be used with an additional NRTI, such as zidovudine or stavudine, to form a complete regimen. Nevirapine binds directly to reverse transcriptase and blocks the RNA-dependent and DNA-dependent DNA polymerase activities, while lamivudine is incorporated into viral DNA by reverse transcriptase, resulting in chain termination.
