Nezastomig + REGN4336 is an investigational combination immunotherapy being developed by Regeneron Pharmaceuticals for the treatment of metastatic castration-resistant prostate cancer (mCRPC). The regimen consists of two bispecific antibodies: nezastomig (REGN5678), which targets PSMA and CD28 to provide costimulatory 'Signal 2' to T-cells, and REGN4336, which targets PSMA and CD3 to provide the primary activation 'Signal 1'. By simultaneously engaging both receptors on T-cells and targeting the prostate-specific membrane antigen (PSMA) on tumor cells, this combination is designed to enhance T-cell proliferation and cytotoxic activity against prostate cancer cells while potentially improving the safety profile compared to high-dose CD3-bispecific monotherapy.
