NG-350A + pembrolizumab is an investigational combination therapy being evaluated for the treatment of metastatic or advanced epithelial tumors. NG-350A is a next-generation, blood-stable, tumor-selective adenoviral vector engineered to express an agonist anti-CD40 antibody. This design enables selective delivery and replication within tumor cells, leading to local expression of the immunostimulatory anti-CD40 antibody and promoting immune-mediated tumor cell killing. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor on T cells, blocking its interaction with PD-L1/PD-L2 and thereby enhancing antitumor immune responses. The combination aims to synergistically activate both innate and adaptive immunity against cancer cells. Clinical trials are ongoing to assess safety, tolerability, pharmacokinetics, pharmacodynamics, and preliminary efficacy in patients with advanced solid tumors[1][3][4][5][6][8].
