Overview

NG-641 + pembrolizumab is an investigational combination therapy for cancer. NG-641 is an oncolytic adenoviral vector engineered to selectively replicate in tumor cells and express multiple transgenes: a fibroblast activation protein-directed bi-specific T-cell activator antibody fragment (FAP-TAc) and an immune enhancer module comprising CXCL9, CXCL10, and interferon alpha2, aiming to target the tumor microenvironment by depleting tumor-associated fibroblasts and recruiting/activating T-cells. Pembrolizumab is a monoclonal antibody targeting PD-1, blocking immune evasion by tumor cells. The combination is under clinical investigation for its potential to synergistically enhance anti-tumor immune responses, specifically in patients with surgically resectable squamous cell carcinoma of the head and neck[1].
